In vitro-Selektion durch Phagendisplay

Die Methode des Phagendisplay ermöglicht es, aus einer sehr großen Bibliothek verschiedener Peptide diejenigen zu selektionieren, die an ein bestimmtes Zielmolekül binden. Dafür werden die Peptide auf Bakteriophagen präsentiert. Die Präsentation von Peptiden auf Phagen (Phagendisplay) wird ermöglicht, indem das Peptid bzw. dessen DNA mit einem Phagenhüllprotein bzw. der dafür kodierenden DNA verknüpft wird. Somit entsteht eine physikalische Verbindung zwischen dem präsentierten Peptid (Phänotyp) und der dafür codierenden DNA (Genotyp). Durch die kombinatorische Mutagenese des Fusionsgens kann eine große Diversität (oft mehr als eine Milliarde verschiedene Varianten)  in das zu präsentierende Peptid eingebracht werden. Durch fortgeschrittene, biotechnologische Methoden können sehr große Anzahlen unterschiedlicher Peptide auf den Phagen präsentiert werden. Anschließend wird durch das sogenannte „Biopanning"-Verfahren die Peptide in der Peptid-Bibliothek angereichert, die besonders stark an ein bestimmtes Zielmolekül binden. Dazu werden die Phagenbibliotheken mit dem Zielmolekül, das auf einer festen Oberfläche immobilisiert wurde, inkubiert. Einige der Peptide (und somit die Phagen) binden stärker als andere an das Zielmolekül. Die weniger stark bindenden werden weggewaschen und die stark bindenden Peptide, bzw. die sie präsentierenden Phagen werden später eluiert, durch Vermehrung in Bakterien amplifiziert und für eine weitere Selektionsrunde benutzt. Somit werden von Selektionsrunde zu Selektionsrunde affinere Bindepartner für das Zielmolekül angereichert.
In diagnostischen oder therapeutischen Ansätzen haben D-enantiomere Peptide verschiedene Vorteile gegenüber natürlich vorkommenden L-enantiomeren Peptiden. Sie sind in vivo proteaseresistent und haben daher häufig eine deutlich erhöhte Halbwertszeit in Blut oder Speichel. Ein sehr eleganter Weg, D-enantiomere Peptide zu identifizieren, ist das Spiegelbild-Phagendisplay. Während bei einem herkömmlichen Phagendisplay L-enantiomere Peptide als Zielmoleküle verwendet werden, werden bei dem Spiegelbild-Phagendisplay D-enantiomere Zielmoleküle benutzt (siehe Abbildung). Somit nutzt das Spiegelbild-Phagendisplay alle Vorteile des üblichen Phagendisplays, nämlich die Selektion neuer Bindepartner aus einer biologischen Bibliothek von bis zu 1013 verschiedener Peptidvarianten. Dies ermöglicht das Schöpfen aus einer riesigen Quelle struktureller Diversität.
