Oberflächenplasmonresonanz (SPR)

Die auf dem gleichnamigen physikalischen Phänomen beruhende Oberflächenplasmonresonanz erlaubt die Detektion von Molekülen bei Bindung an eine Oberfläche.

Im Falle der Totalreflektion von eingestrahltem Licht wird ein sogenanntes evaneszentes Feld generiert, das eine begrenzte Eindringtiefe von ~300 nm besitzt. Sind die Resonanzbedingungen erfüllt, so ist das evaneszente Feld in der Lage mit Oberflächenplasmonen eines dünnen Metallfilms zu interagieren. Dabei sind die Resonanzbedingungen abhängig von folgenden Parametern: (i) Einfallwinkel, (ii) Brechungsindizes und (iii) Wellenlänge. Während eines Experiments wird die Wellenlänge konstant gehalten. So lassen sich Veränderungen des Brechungsindex in der Nähe der Oberfläche des Metallfilms durch die Veränderung des Resonanzwinkels detektieren. Da die Bindung von Molekülen and die Oberfläche zu einer Veränderung des Brechungsindex führt, lässt sich die Bindung von Biomolekülen zeitaufgelöst messen und darstellen.

Das resultierende Sensorgramm ist eine Darstellung des Oberflächenplasmonenresonanzsignals in Resonanzeinheiten (RU) gegen die Zeit (t). Hierbei entspricht eine Resonanzeinheit der Bindung von 1 pg an die Oberfläche einer Flusszelle mit der Fläche von 1 mm2. Demnach ist die Einheit RU definiert als pg/mm2
