Viroide

Viroide sind zirkuläre, einzelsträngige, nicht-kodierende RNAs, die in der Lage sind bestimmte Pflanzen zu infizieren. Da Viroide für kein Peptid oder Protein kodieren, müssen sie Wirtsproteine für ihre biologische Funktion benutzen; dazu zählen zum Beispiel Replikation, Prozessierung und Transport. Daher kann man Viroide als minimale Parasiten der Wirtsmaschinerie betrachten.  Dazu müssen Viroide der Wirtsmaschinerie die entsprechenden Signale präsentieren, die entweder in  ihrer Sequenz oder  in ihrer Struktur kodiert sein müssen.

Das Kartoffel-Spindelknollensucht-Viroid (potato spindle tuber viroid, PSTVd) ist namensgebend für die Viroid-Familie Pospiviroidae. Der Replikationszyklus von PSTVd ist ein asymmetrischer "rolling circle"-Mechanismus (siehe linke Abbildung). Unter nativen Bedingungen besitzt das maturierte, zirkuläre, (+)-strängige Viroid (oben in der Abbildung) eine stäbchenförmige Sekundärstruktur aus kurzen Helices und einzelsträngigen Bereichen (Loops). Diese Struktur ist für die partielle Resistenz von Viroiden gegen Nukleasen verantwortlich. Die zirkuläre, (+)-strängige RNA wird durch die DNA-abhängige RNA-Polymerase II (polII) der Wirtspflanze in einen oligomeren (-)-Strang transkribiert (rechts in der Abbildung). Während der Transkription faltet sich die (-)-strängige RNA in eine thermodynamisch metastabile Konformation, die die Haarnadelschleife II (hairpin II, HPII) enthält. Diese ist wichtig für die Transkription der (-)-strängigen Oligomere in (+)-Stränge durch die polII (unten in der Abbildung). Die (+)-strängigen Oligomere, deren 5'- und 3'-Enden in der Abbildung blau markiert sind, werden durch Wirtsenzyme an einem einzigartigen Strukturelement, das eine thermodynamisch besonders stabile, vier Nukleotide lange Haarnadelschleife (Tetraloop, tl) und ein G:U-Basenpaar enthält, erkannt, zu Monomeren geschnitten und zu Zirkeln ligiert.

Vom maturierten PSTVd wird angenommen, dass es für den intrazellulären und systemischen Transport entscheidend ist. PSTVd-Stämme, die sich durch wenige Mutationen im linken Teil ihrer Sekundärstruktur unterscheiden (bezeichnet mit PM für pathogenitätsmodulierende Region in obiger Abbildung), verursachen in der Wirtspflanze Symptome, die von geringfügig (mild) bis drastisch ("lethal") reichen können. In der linken Abbildung sind Tomatenpflanzen gleichen Alters und gleicher Dauer einer PSTVd-Infektion gezeigt: Die linke Pflanze ist schein-infiziert (mock); die weiteren wurden mit einem milden Stamm (QFA), einem intermediären bzw. einem starke Symptome auslösenden PSTVd-Stamm (AS1) infiziert. Neben dem üblichen gestauchten Habitus der Viroid-infizierten Pflanzen verursacht PSTVd-AS1 Symptome bis zur Nekrose von Stängel und Blättern.

Der molekulare Mechanismus der Viroid-induzierten Pathogenese ist bisher ungeklärt. Das aktuelle Pathogenitätsmodell favorisiert die Hypothese, dass Viroide als Quelle für "silencing" oder microRNAs dienen; dies sind kurze RNAs mit Viroidsequenz und einer Länge von 21 bis 24 Nukleotiden. Die kurzen RNAs könnten in der Lage sein, mit mit der Genregulation des Wirts zum Beispiel durch "transcriptional" oder "post-transcriptional gene silencing" (TGS oder PTGS) zu interferieren.

Die Analyse der PSTVd-induzierten kleinen RNAs und ihres Zusammenhangs mit Viroid-Pathogenität wird gemeinsam von den Arbeitsgruppen G. Steger und J. Matoušek (undefinedBiologisches Zentrum, Akademie der Wissenschaften der Tschechischen Republik) untersucht. Diese Arbeiten werden zur Zeit von der undefinedAlexander-von-Humboldt-Stiftung und durch die Graduierten-Schule undefinedE-Norm im Projekt "Viroid-Evolution und Viroid-induzierte Pathogenese-Netzwerke in Wirtspflanzen" unterstützt. Von G. Steger und J. Matoušek wurde die undefined"International Conference on Viroids and Viroid-like RNA" (2015) organisiert.
